Correlation between N-Acetyltransferase 2 (NAT2) Polymorphism Genotype with Plasma Isoniazid (INH) Concentration in MDR TB Patients Receiving Short Regimen in West Sumatera

Background: Isoniazid (INH) is one of the most potent TB drug. High dose INH used in short regimen MDR drugs. The genetic polymorphism NAT2 affects acetylation status. Awareness patients’ acetylator status important to determine risk toxicity, treatment failure and drug resistance. aim this study was demonstrate genotype association with plasma concentration after 2 hours oral therapy. Methods: This a cross sectional patients who received term combination therapy at RSUP Dr. M.Djamil Padang, Achmad Muchtar Hospital Bukittinggiand West Sumatra Pulmonary from September 2019 February 2020. Patients were examined for concentration. results concentrations obtained evaluated based on phenotype group.Results: majority subjects weremen (62.5%), aged 40-64 years (50%), had common comorbid diabetes mellitus (31.25%), normoweight (75%) negative HIV (93.8%). A total 7 alleles consisting SNPs variations found short-term NAT2*12A (56.25%) allele fast acetylator. Based bimodal distribution, median slow 1.25 µg/ml 5.24 µg/ml, respectively. values trimodal distribution fast, intermediate, acetylators 2.17 µg/ml. Conclusion: There no correlations between type concentrations.
